Does FOXO4-DRI help with senescent cell clearance?

D
Grade D — no human evidenceCell culture · effect size: moderate
Short answerNo human evidence. As of 23 September 2026, only cell culture exist for FOXO4-DRI for senescent cell clearance. Removed senescent cells from expanded human chondrocytes in a 2021 study — the closest the compound has come to a human tissue.

Animal studies, cell studies, or anecdotes only. This is where almost every viral peptide sits. Promising and proven are different words for a reason.

In context

FOXO4-DRI is a cell-penetrating peptide designed to pry the protein FOXO4 away from p53 inside senescent cells, causing those cells to die. In a 2017 Cell paper it restored fitness, fur density and kidney function in aged and prematurely ageing mice. That is the entire evidence base: no human trial, no human safety data, and no regulatory filing. Products sold under the name are unlicensed research chemicals.

FOXO4-DRI is not approved (preclinical). Legal status differs by country — see the regulation section or the jurisdiction pages.

Questions

Is FOXO4-DRI proven to help with senescent cell clearance?
No. There is no human evidence for it (Cell culture); the claim rests on animal or cell studies and LYO grades it D.
What studies are behind the grade?
Removed senescent cells from expanded human chondrocytes in a 2021 study — the closest the compound has come to a human tissue.
Is FOXO4-DRI approved for this?
No. FOXO4-DRI is not approved (preclinical); it is not an approved medicine for any indication in the jurisdictions LYO covers.
