Does Matrixyl help with collagen synthesis?

D
Grade D — no human evidenceNone in vivo · effect size: not measurable
Short answerNo human evidence. As of 29 October 2026, only none in vivo exist for Matrixyl for collagen synthesis. The mechanism rests on fibroblast culture work showing that KTTKS stimulates collagen I, collagen III and fibronectin production. No human study has measured collagen synthesis in skin after topical pal-KTTKS.

What the studies show

The mechanism rests on fibroblast culture work showing that KTTKS stimulates collagen I, collagen III and fibronectin production. No human study has measured collagen synthesis in skin after topical pal-KTTKS.

Animal studies, cell studies, or anecdotes only. This is where almost every viral peptide sits. Promising and proven are different words for a reason.

In context

Matrixyl is palmitoyl pentapeptide-4 (pal-KTTKS), a five-amino-acid fragment of type I collagen with a palmitic acid tail added so it can cross the skin barrier. Its human evidence is a 93-woman, 12-week, split-face randomised trial run by Procter & Gamble, which found significant reduction in wrinkles and fine lines against the vehicle control by both instrumental and expert-grader analysis. It is a legal cosmetic ingredient everywhere LYO covers and is not a medicine.

Matrixyl is cosmetic ingredient. Legal status differs by country — see the regulation section or the jurisdiction pages.

Questions

Is Matrixyl proven to help with collagen synthesis?
No. There is no human evidence for it (None in vivo); the claim rests on animal or cell studies and LYO grades it D.
What studies are behind the grade?
The mechanism rests on fibroblast culture work showing that KTTKS stimulates collagen I, collagen III and fibronectin production. No human study has measured collagen synthesis in skin after topical pal-KTTKS.
Is Matrixyl approved for this?
No. Matrixyl is cosmetic ingredient; it is not an approved medicine for any indication in the jurisdictions LYO covers.
